I don't get it.
Director's Commentary: A meta-joke is a joke about a joke or jokes, but more commonly about a category of jokes. In this scene Duke Nukem is telling a meta-joke about meta-jokes, ergo this scene is hilarious because of his hot pink vest.
I feel like I have both learned something and suffered minor brain damage all at once!
